The COT n/a (Catalog #MBS641654) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The COT, CT (Cancer Osaka Thyroid Oncogene, c-COT, EST, ESTF, Mitogen-activated Protein Kinase Kinase Kinase 8, MAP3K8, MEKK8, Proto-oncogene c-Cot, Serine/threonine-protein Kinase Cot, Tumor Progression Locus 2, TPL2, TPL-2) reacts with Human and may cross-react with other species as described in the data sheet. MyBioSource\'s COT can be used in a range of immunoassay formats including, but not limited to, ELISA (EL/EIA), Western Blot (WB), Immunohistochemistry (IHC).Suitable for use in ELISA, Western Blot, and Immunohistochemistry.
Dilution: ELISA: 1:1,000
Western Blot: 1:100-1:500
Immunohistochemistry: 1:50-1:100. Researchers should empirically determine the suitability of the COT n/a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.

Please refer to the product datasheet for known applications of a given antibody. We\'ve tested the COT, CT (Cancer Osaka Thyroid Oncogene, c-COT, EST, ESTF, Mitogen-activated Protein Kinase Kinase Kinase 8, MAP3K8, MEKK8, Proto-oncogene c-Cot, Serine/threonine-protein Kinase Cot, Tumor Progression Locus 2, TPL2, TPL-2) with the following immunoassay(s):
Western Blot (WB) (Western Blot to detect COT in HeLa cell lysate using MBS641654.)
Immunohistochemistry (IHC) (Immunohistochemistry analysis in formalin-fixed and paraffin-embedded human cancer tissue reacted with MBS641654, which was peroxidase-conjugated to the secondary antibody, followed by DAB staining. This data demonstrates the use of MBS641654 for immunohistochemistry. BC = breast carcinoma; HC = hepatocarcinoma.)
Testing Data
COT, a member of the MAPKKK subfamily of Ser/Thr protein kinases, is able to activate NF-kappa-B 1 by stimulating proteasome-mediated proteolysis of NF-kappa-B 1/p105. It plays a role in the cell cycle. The longer form of cot has some transforming activity, although it is much weaker than the activated cot oncoprotein. This cytoplasmic protein is expressed in several normal tissues and human tumor-derived cell lines. The 58kD form is activated specifically during the S and G2/M phases of the cell cycle. The longer form undergoes phosphorylation on Ser residues mainly, and the shorter form on both Ser and Thr residues.
Immunogen: Synthetic peptide selected from the C-terminal region of human COT (KLH). In general, we may offer more than one antibody to a given target to enable options for the researcher. Available antibodies recognizing COT are readily searchable from our website. Different antibodies against the same target such as COT may be optimized or tested for different applications and species. This enables researchers to select the option that may be best for their model system, to screen more than antibody to determine which one may be best for their model system, as well as to use more than one antibody to follow up on and validate their results.
