The Histone H3, dimethyl (Lys27) reacts with Human and may cross-react with other species as described in the data sheet. MyBioSource\'s Histone H3, dimethyl can be used in a range of immunoassay formats including, but not limited to, Western Blot (WB), Immunoprecipitation (IP), Immunocytochemistry (ICC), Gel Shift Assay (GS/EMSA).Suitable for use in Western Blot, Peptide Inhibition, Immunocytochemistry, Chromatin Immunoprecipitation, Beadlyte Histone-Peptide Specificity Assay and Dot Blot.
Dilution: Western Blot: 0.5ug/ml detects dimethylated histone H3 in acid extracted proteins from HeLa cells. No detection of 1ug unmethylated recombinant Histone H3 (H5110-12K) was observed.
Peptide Inhibition: 1uM of the immunizing peptide to completely abolishes detection of histone H3 in Western Blot analysis of HeLa acid extracts. Slight signal reduction was observed with peptide containing dimethyl-lysine 9. Signal was not reduced by branched monomethyl H3(Lys27), linear monomethyl H3(Lys27), linear trimethylH3(Lys27), or linear dimethyl H3(Lys4) peptides.
Beadlyte Histone-Peptide Specificity Assay: 12.5ng/ml or a range of dilutions detected peptides containing the dimethyl-lysine modification at residue 27. Peptides containing other modifications were not detected. Researchers should empirically determine the suitability of the H3 n/a for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.

Chemiluminescence (Recombinant Histone H3 (H5110-14P) (Lane 1) and HeLa acid extract (Lane 2) were resolved by electrophoresis, transferred to PVDF and probed with MBS618360 (0.5ug/ml). Proteins were visualized using a donkey anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Arrow indicates Histone H3 at ~17kD.)
Testing Data (12.5ng/ml (Figure B) of MBS618360 was incubated with a cocktail of microspheres conjugated to branched histone H3 peptides with the following modifications: 1. dimethyl-Histone H3 (Lys9) 2. monomethyl-Histone H3 (Lys27) 3. triimethyl-Histone H3 (Lys27) 4. dimethyl-Histone H3 (Lys27).)

Histone H3 is one of the 5 main histone proteins involved in the structure of chromatin in eukaryotic cells. Featuring a main globular domain and a long N terminal tail H3 is involved with the structure of the nucleosomes of the \'beads on a string\' structure. Acetylation of histone H3 occurs at several different lysine positions in the histone tail and is performed by a family of enzymes known as Histone Acetyl Transferases (HATs).
