LAG-3, also known as CD223, is a type I membrane protein that belongs to the immunoglobulin superfamily (IGSF). The extracellular domain of LAG-3 contains a V-set and three C2-set Ig domains. LAG-3 is expressed on activated T cells, NK cells and B cells. It binds to MHC class II molecules with much higher avidity than CD4, a LAG-3 structural homolog. Mouse and human LAG-3 have been shown to be negative regulators of T cell function and homeostasis. The extracellular domain of human and mouse LAG-3 share 69% aa sequence identity.
