Neuropilin-2 (np-2, Npn-2, NRP2) belongs to a small family of Type I transmembrane receptors that also contains the structurally similar neuropilin-1. Neuropilins were first identified as mediators of Class III semaphorins in axonal guidance; neuropilin-2 acts as a receptor for semaphorin-3A and -3F, and for specific splice variants of VEGF, including VEGF165 and VEGF145.1 Although they share 44% amino acid identity, neuropilin-1 and -2 differ structurally in their transmembrane and cytoplasmic domains,2 as well as in tissue distribution patterns. Whereas neuropilin-1 is expressed in the heart and capillaries, neuropilin-2 is absent there, and present in the dorsal aorta colliculus instead.1 In the rat uterus, neuropilin-1 was identified in both endothelial and glandular cells, while Npn-2 was present only in the glandular cells, not the vasculature.3 Another location where neuropilin-2 is expressed is in neuroendocrine cells of the human digestive tract. By confocal microscopy, Npn-2 was observed in vesicle-like structures near the nucleus at the basolateral side of cells.4 Interestingly, gastrointestinal carcinoid tumors of the colon and appendix, which develop from neuroendocrine cells, do not contain neuropilin-2-producing cells.4 This finding suggests that loss of Npn-2 expression may accompany tumor progression.
