Phosphoinositide-specific phospholipase C (PLC) is a family of proteins that hydrolyze phosphatidylinositol 4,5-bisphosphate to generate the second messengers, inositol 1,4,5-trisphosphate (IP3) and diacylglycerol (DAG). IP3 induces a transient increase in intracellular free Ca++, while DAG is a direct activator of protein kinase C (PKC). These processes are involved in the control of multiple cellular activities such as cell proliferation, growth and differentiation. PLCy-1 is a 135kD ubiquitously expressed isozyme that is activated by phosphorylation of tyrosine 783 and interacts with growth factor receptors (EGFR, PDGFR, etc.) and adhesion/cytoskeletal proteins (FAK, paxillin) to influence a multiple number of signaling pathways.
