The TH th (Catalog #MBS221802) is an Antibody produced from Rabbit and is intended for research purposes only. The product is available for immediate purchase. The RABBIT ANTI TYROSINE HYDROXYLASE (pSer40) reacts with Human (Expected from Sequence), Mouse (Expected from Sequence), Pig (Expected from Sequence), Rat (Expected from Sequence) and may cross-react with other species as described in the data sheet. MyBioSource\'s TYROSINE HYDROXYLASE can be used in a range of immunoassay formats including, but not limited to, Immunohistology Frozen, Immunofluorescence (IF), Western Blot (WB)*.Western Blot: This item detects a band of approximately 60kDa in PC-12 cell lysates, following stimulation by Okadaic acid.
Immunohistology - Frozen: Maximum Dilution: 1/1000
Immunofluorescence: Maximum Dilution: 1/1000
Western Blotting: Maximum Dilution: 1/1000; Application Note: For the detection of phosphoproteins, serine phosphatase inhibitors such as 10mM Sodium Fluoride should be added to the sample buffer. Milk or other casein-based blocking solutions are not recommended as casein is a phosphoprotein and its use can result in high background. Researchers should empirically determine the suitability of the TH th for an application not listed in the data sheet. Researchers commonly develop new applications and it is an integral, important part of the investigative research process.
The TH th product has the following accession number(s) (GI #88900503) (NCBI Accession #NP_000351.2) (Uniprot Accession #P07101). Researchers may be interested in using Bioinformatics databases such as those available at The National Center for Biotechnology Information (NCBI) website for more information about accession numbers and the proteins they represent. Even researchers unfamiliar with bioinformatics databases will find the NCBI databases to be quite user friendly and useful.

MBS221802 is specific for Tyrosine Hydroxylase, when phosphorylated at serine 40. Tyrosine hydroxylase (TH) catalyzes the rate-limiting step in the biosynthetic pathway of the catecholamines dopamine (DA), norepinephrine, and epinephrine. The enzyme exists as a tetramer, with each subunit composed of an N-terminal regulatory domain and a C-terminal catalytic domain. Phosphorylation of TH has been shown to occur at several serine residues. Phosphorylation at serine 40 results in an increase in hydroxylase activity, and phosphorylation at serine 19 is reported to promote phosphorylation of the serine 40 residue. Tyrosine hydroxylase is regularly used as a marker for dopaminergic neurons, which is particularly relevant for research into Parkinson\'s disease.
Perservative Stabilisers: 0.09% Sodium Azide
0.01% Bovine Serum Albumin
50% Glycerol. Immunogen: Synthetic phosphopeptide corresponding to an amino acid sequence within Tyrosine Hydroxylase which includes phosphorylated Ser 40.
Buffer Solution: 10mM HEPES pH7.5
Antiserum Preparation: Antisera to phosphorylated tyrosine hydroxylase were raised by repeated immunisations of rabbits with highly purified antigen. Purified IgG prepared by affinity chromatography. In general, we may offer more than one antibody to a given target to enable options for the researcher. Available antibodies recognizing TH are readily searchable from our website. Different antibodies against the same target such as TH may be optimized or tested for different applications and species. This enables researchers to select the option that may be best for their model system, to screen more than antibody to determine which one may be best for their model system, as well as to use more than one antibody to follow up on and validate their results.
